Salem band marches in Fiesta Bowl parade

Members of the Salem High School marching band and color guard got a huge thrill Saturday.

The Blue Devils marched in the annual Fiesta Bowl Parade in Phoenix, Ariz. The band played the song "The Battle Hymn of the Republic."

This was not the first time the Salem band has played at the Fiesta Bowl. One of the current assistant directors met her husband at the parade in 2004.

NHL, union spend day on conference calls

NEW YORK (AP) ? Lots of questions, but still no answers in the NHL labor fight.

The league and the players' association spent much of Saturday talking to each other via conference call. The conversations were strictly for the purpose of sharing information regarding the new contract offer the NHL made to the union late Thursday.

The initial thought was that the sides would get together Sunday in New York to hold official negotiations for the first time in weeks, but those weren't scheduled before discussions ended Saturday.

Staff level calls were expected to resume Sunday, which could include face-to-face talks at some point, but no actual bargaining meetings have been set.

"Some more informational sessions in the morning," NHL deputy commissioner Bill Daly told The Associated Press regarding Sunday. "Nothing planned beyond that."

The union pored over the latest NHL proposal aimed at ending the lockout that reached its 105th day Saturday and saving the delayed hockey season. The league would like to reach a deal no later than Jan. 11, which would allow training camps to start the following day, and a 48-game season to begin Jan. 19.

Saturday's conference calls were scheduled Friday night so the union could ask league officials questions about the nearly 300-page proposal. Whether enough progress will be made to lead to face-to-face talks remains to be seen.

The sides haven't gotten together since Dec. 13 with federal mediators. Bargaining sessions with only the NHL and union involved haven't been held since Dec. 6, when talks abruptly ended after the players' association made a counterproposal to the league's previous offer. The league said that offer was contingent on the union accepting three elements unconditionally and without further bargaining.

The NHL then pulled all existing offers off the table. Two days of sessions with mediators the following week ended without any progress made.

The players' association's executive board and negotiating committee went over the new proposal during an internal conference call Friday.

A person familiar with key points of the offer told The Associated Press that the league proposed raising the limit of individual free-agent contracts to six years from five ? seven years if a team re-signs its own player; raising the salary variance from one year to another to 10 percent, up from 5 percent; and one compliance buyout for the 2013-14 season that wouldn't count toward a team's salary cap but would be included in the overall players' share of income.

The person spoke on condition of anonymity because details of the new offer weren't being discussed publicly.

The NHL maintained the deferred payment amount of $300 million it offered in its previous proposal, an increase from an earlier offer of $211 million. The initial $300 million offer was pulled off the table after negotiations broke off earlier this month.

The latest proposal is for 10 years, running through the 2021-22 season, with both sides having the right to opt out after eight years.

The lockout has reached a critical stage, threatening to shut down a season for the second time in eight years. All games through Jan. 14, plus the Winter Classic and the All-Star game already have been called off. The next round of cuts could claim the entire schedule.

The NHL is the only North American professional sports league to cancel a season because of a labor dispute, losing the 2004-05 campaign to a lockout. A 48-game season was played in 1995 after a lockout stretched into January.

It is still possible this dispute could eventually be settled in the courts if the sides can't reach a deal on their own.

The NHL filed a class-action suit this month in U.S. District Court in New York in an effort to show its lockout is legal. In a separate move, the league filed an unfair labor practice charge with the National Labor Relations Board, contending bad-faith bargaining by the union.

Those moves were made because the players' association took steps toward potentially filing a "disclaimer of interest," which would dissolve the union and make it a trade association. That would allow players to file antitrust lawsuits against the NHL.

Union members voted overwhelmingly to give their board the power to file the disclaimer by Jan. 2. If that deadline passes, another authorization vote could be held to approve a later filing.

Negotiations between the NHL and the union have been at a standstill since talks ended Dec. 6. One week later, the sides convened again with federal mediators in New Jersey, but still couldn't make progress.

The sides have been unable to reach agreement on the length of the new deal, the length of individual player contracts, and the variance in salary from year to year. The NHL is looking for an even split of revenues with players.

WASHINGTON (AP) ? When it comes to the nation's budget challenges, congressional leaders are fond of saying dismissively they don't want to kick the can down the road.

But now, a deadline hard ahead, even derided half-measures are uncertain as President Barack Obama and lawmakers struggle to avert across-the-board tax increases and spending cuts that comprise an economy-threatening fiscal cliff.

Congressional officials said Wednesday they knew of no significant strides toward a compromise over a long Christmas weekend, and no negotiations have been set.

After conferring on a conference call, the House Republican leadership said they remain ready for talks, but gave no hint they intend to call lawmakers back into session unless the Senate first passes legislation.

"The lines of communication remain open, and we will continue to work with our colleagues to avert the largest tax hike in American history, and to address the underlying problem, which is spending," the leadership said in a statement.

The Senate is due in session Thursday, although the immediate agenda includes legislation setting the rules for government surveillance of suspected spies and terrorists abroad, including Americans, as well as a measure providing $60 billion for victims of Superstorm Sandy.

Obama decided to cut short his Hawaii vacation for an overnight flight expected to get him back to the White House on Thursday.

Apart from the cliff, other financial challenges loom for divided government, where political brinkmanship has become the norm. "We have a spending problem. We have to address it, And we're not going to address it by kicking the can down the road," the speaker said at a news conference late last week when he was asked about setting a vote on a plan that Democrats find acceptable.

In July 2011, when he was struggling with Republicans over the threat of a first-ever government default, Obama said he had "heard reports that there may be some in Congress who want to do just enough to make sure that America avoids defaulting on our debt in the short term. But then wants to kick the can down the road when it comes to solving the larger problem, our deficit."

A few months later, an extension of a payroll tax cut was the issue, and Boehner was insisting on a year-long renewal rather than the temporary plan that passed the Senate with votes from lawmakers in both parties.

"How can you do tax policy for two months?" he asked on Dec. 18, 2011. "I believe that two months is just kicking the can down the road.

"The American people are tired of that."

At issue now is series of tax increases and spending cuts scheduled to kick in with the new year that economists caution could send the economy into a recession.

India, Pakistan Cricket: Bitter Rivals Face Off In Bangalore In First Bilateral Series Since 2007

BANGALORE, India ? Four years after Pakistani gunmen laid siege to India's financial capital of Mumbai, South Asia's bitter rivals were meeting again on the cricket ground, marking a gradual thaw in their decades-old rivalry.

The first bilateral series between India and Pakistan since November 2007, comprising two Twenty20 matches and three one-day internationals, began Christmas Day with a Twenty20 match in the southern Indian city of Bangalore.

Pakistan won the match with five wickets to spare after India's batting collapsed at 133 for nine.

Thousands of cricket fans began lining up outside Bangalore's massive Chinnaswamy Stadium nearly five hours before the match was to begin.

"This match is like no other. There's a special thrill to a match where India faces Pakistan," said Ravinder Singh, his loyalties evident from the Indian flag colors painted on his cheeks.

"I'm telling my friends it will be worth the wait," said Singh, a college student, as he stood in a slow-moving line outside the stadium. Some of his friends were in the sky blue shirts of the Indian team.

Security was tight with thousands of paramilitary soldiers and police outside the stadium. Groups of police carried out body searches before allowing fans into the stadium after they had gone through metal detectors.

Unflustered by the tight security, cricket fans carried flags and pro-India banners while a few sported colorful wigs and face-paint.

Analysts see the cricket series as a sign the two sides are ready to move past the bitterness that followed the 2008 Mumbai terror attacks, when 10 Pakistan-based gunmen killed 166 people in a three-day rampage across the city.

India blamed the Lashkar-e-Taiba militant group for the attacks and demanded that Islamabad crack down on terrorism.

Despite a long history of mutual distrust and animosity, the love of cricket ? bequeathed to India and Pakistan by South Asia's British colonial rulers ? is one of the few things the countries agree upon.

Relations have improved since the Mumbai attacks and diplomatic ties have been renewed, but New Delhi remains unsatisfied with the slow pace of Islamabad's efforts to bring the perpetrators of the attacks to justice.

New Delhi froze nearly all contact with Islamabad ? including sporting ties ? after the Mumbai attacks, a hiatus that has been bridged in recent years by India and Pakistan playing matches in third countries or in international meets such as the World Cup.

In the years since the Mumbai attacks, some efforts have been made to bring bilateral relations out of the deep freeze. Direct trade has been increasing steadily as both countries make efforts to increase trade across their land border.

At the Wagah-Attari land border in Punjab, India has opened a huge customs depot and warehouses that can handle more than 600 trucks a day from Pakistan. Two-way trade direct between India and Pakistan totals around $2 billion, but a large chunk of the trade is channeled through Dubai, Hong Kong or Singapore.

Earlier this month, India and Pakistan signed an agreement that makes it easier for business travelers to get visas. People aged over 65 also will be entitled to get visas on arrival. Members of families divided during Britain's partition of the subcontinent, along with tourists and religious pilgrims, are also supposed to get quick visas.

"When Indians enter Pakistan and when Pakistanis enter India, they should feel like they are coming home," Rehman Malik, Pakistan's interior minister, said in New Delhi two weeks ago when the visa agreement was signed. India has issued more than 3,000 visas to Pakistanis for the cricket matches.

But analysts caution that policy makers in India should not get carried away by the friendly neighbor rhetoric.

"All forms of people-to-people contact, including sports, are important and should be pursued, but never at the cost of our main focus, which is terrorism emanating from Pakistan," said Vivek Katju, a retired diplomat who has served in Pakistan and was India's ambassador in Afghanistan.

Across the border, Pakistani analysts feel that while the resumption of sporting contact is welcome, the two can make real progress only when they succeed in resolving their long-standing disputes.

Rasool Bakhsh Rais, a professor of political science at the Lahore University of Management Sciences in Pakistan, said sports could be a "major avenue through which hostilities between the two nations could be set aside."

The expectations riding on cricket players are huge before any match, but especially when they play against their great rivals. So great are the pressures, a sports psychologist is accompanying the Pakistani team during its stint in India, Pakistani cricket officials said.

In Forney?s Diamond Creek neighborhood, though, families are still trickling back, eight months after a tornado struck.

Some homes, being rebuilt from scratch, remain under construction. Others have disappeared entirely, leaving just a slab or empty lot behind. Several appear virtually untouched since the tornado, with broken windows and gaping holes.

?It was surreal. It was a warzone,? said Ronnie Smith.

The father of two remembers running past collapsed houses, searching for his own. ?You know that feeling where you just kind of lose your breath and you can?t get your breath back again?? he said.

His house escaped the worst of the storm, but still required repairs, that forced the family to stay with relatives for three months.

It?s only now they?re starting to feel settled again. ?My wife told me yesterday that our house is just now becoming a home again,? he said.

Monica McCrea said, her family couldn?t wait to come back, and returned home on Labor Day. ?Being in your own home is a wonderful thing,? she said.

The neighborhood is lot quieter now, and there aren?t as many Christmas decorations this year.
Those who?ve returned, though, say neighbors feel closer than ever.

WASHINGTON (AP) ? A U.S. Army brigade will begin sending small teams into as many as 35 African nations early next year, part of an intensifying Pentagon effort to train countries to battle extremists and give the U.S. a ready and trained force to dispatch to Africa if crises requiring the U.S. military emerge.

The teams will be limited to training and equipping efforts, and will not be permitted to conduct military operations without specific, additional approvals from the secretary of defense.

The sharper focus on Africa by the U.S. comes against a backdrop of widespread insurgent violence across North Africa, and as the African Union and other nations discuss military intervention in northern Mali.

The terror threat from al-Qaida linked groups in Africa has been growing steadily, particularly with the rise of the extremist Islamist sect Boko Haram in Nigeria. Officials also believe that the Sept. 11 attack on the U.S. consulate in Benghazi, which killed the ambassador and three other Americans, may have been carried out by those who had ties to al-Qaida in the Islamic Maghreb.

This first-of-its-kind brigade assignment ? involving teams from the 2nd Brigade, 1st Infantry Division ? will target countries such as Libya, Sudan, Algeria and Niger, where al-Qaida-linked groups have been active. It also will assist nations like Kenya and Uganda that have been battling al-Shabab militants on the front lines in Somalia.

Gen. Carter Ham, the top U.S. commander in Africa, noted that the brigade has a small drone capability that could be useful in Africa. But he also acknowledged that he would need special permission to tap it for that kind of mission.

"If they want them for (military) operations, the brigade is our first sourcing solution because they're prepared," said Gen. David Rodriguez, the head of U.S. Army Forces Command. "But that has to go back to the secretary of defense to get an execute order."

Already the U.S. military has plans for nearly 100 different exercises, training programs and other activities across the widely diverse continent. But the new program faces significant cultural and language challenges, as well as nagging questions about how many of the lower-level enlisted members of the brigade, based in Fort Riley, Kan., will participate, since the teams would largely be made up of more senior enlisted troops and officers. A full brigade numbers about 3,500, but the teams could range from just a few people to a company of about 200. In rare cases for certain exercises, it could be a battalion, which would number about 800.

To bridge the cultural gaps with the African militaries, the Army is reaching out across the services, the embassies and a network of professional organizations to find troops and experts that are from some of the African countries. The experts can be used during training, and the troops can both advise or travel with the teams as they begin the program.

"In a very short time frame we can only teach basic phrases," said Col. Matthew McKenna, commander of the 162nd Infantry Brigade that will begin training the Fort Riley soldiers in March for their African deployment. "We focus on culture and the cultural impact ? how it impacts the African countries' military and their operations."

Thomas Dempsey, a professor with the Africa Center for Strategic Studies, said the biggest challenge will be the level of cultural, language and historical diversity across the far-flung continent.

"How do you train for that in a way that would be applicable wherever they go?" said Dempsey, a retired Army colonel. He said he's not sure using a combat brigade is the right answer, but added, "I'm not sure what the answer is. The security challenges differ so dramatically that, to be honest, I really don't think it's feasible to have a continental training package."

The Pentagon's effort in Africa, including the creation of U.S. Africa Command in 2007, has been carefully calibrated, largely due to broad misgivings across the continent that it could spawn American bases or create the perception of an undue U.S. military influence there. As a result, the command has been based in Stuttgart, Germany, rather than on the African continent.

At the same time, many African nations are eager for U.S. training or support, as they work to build their militaries, battle pirates along the coast and shut down drug trafficking, kidnapping and other insurgent activities.

McKenna acknowledged the challenge, but said the military has to tap its conventional fighting forces for this task because there aren't enough special operations forces to meet the global training needs. He said there will be as many as a dozen different training segments between February and September, each designed to provide tailored instruction for the particular teams.

The mission for the 2nd Brigade ? known as the "Dagger Brigade" ? will begin in the spring and will pave the way for Army brigades to be assigned next to U.S. Pacific Command and then to U.S. European Command over the next year. The brigade is receiving its regular combat training first, and then will move on to the more specific instruction needed for the deployments, such as language skills, cultural information and other data about the African nations.

Dagger Brigade commander Col. Jeff Broadwater said the language and culture training will be different than what most soldiers have had in recent years, since they have focused on Pashtun and Farsi, languages used mostly in Afghanistan and Iran. He said he expects the soldiers to learn French, Swahili, Arabic or other languages, as well as the local cultures.

"What's really exciting is we get to focus on a different part of the world and maintain our core combat skills," Broadwater said, adding that the soldiers know what to expect. "You see those threats (in Africa) in the news all the time."

The brigade will be carved up into different teams designed to meet the specific needs of each African nation. As the year goes on, the teams will travel from Fort Riley to those nations ? all while trying to avoid any appearance of a large U.S. military footprint.

"The challenge we have is to always understand the system in their country," said Rodriguez, who has been nominated to be the next head of Africa Command. "We're not there to show them our system, we're there to make their system work. Here is what their army looks like, and here is what we need to prepare them to do."

Rodriguez said the nearly 100 assignments so far requested by Ham will be carried out with "a very small footprint to get the high payoff."

Israel rejects US gun lobby claims on its security

JERUSALEM (AP) ? Israel's policy on issuing guns is restrictive, and armed guards at its schools are meant to stop terrorists, not crazed or disgruntled gunmen, experts said Monday, rejecting claims by America's top gun lobby that Israel serves as proof for its philosophy that the U.S. needs more weapons, not fewer.

Far from the image of a heavily armed population where ordinary people have their own arsenals to repel attackers, Israel allows its people to acquire firearms only if they can prove their professions or places of residence put them in danger. The country relies on its security services, not armed citizens, to prevent terror attacks.

Though military service in Israel is compulsory, routine familiarity with weapons does not carry over into civilian life. Israel has far fewer private weapons per capita than the U.S., and while there have been gangster shootouts on the streets from time to time, gun rampages outside the context of the Israeli-Palestinian conflict are unheard of.

The National Rifle Association responded to the Dec. 14 killing of 20 first-graders and six adults at a Connecticut elementary school by resisting calls for tighter gun control and calling for armed guards and police at schools. On Sunday, the lobby's chief executive, Wayne LaPierre, invoked his perception of the Israeli school security system to back his proposal.

"Israel had a whole lot of school shootings until they did one thing: They said, 'We're going to stop it,' and they put armed security in every school and they have not had a problem since then," LaPierre said on the NBC News show "Meet the Press."

Israel never had "a whole lot of school shootings." Authorities could only recall two in the past four decades.

In 1974, 22 children and three adults were killed in a Palestinian attack on an elementary school in Maalot, near the border with Lebanon. The attackers' goal was to take the children hostage and trade them for imprisoned militants.

In 2008, another Palestinian assailant killed eight young people, most of them teens, at a nighttime study session at a Jewish religious seminary in Jerusalem. An off-duty soldier who happened to be in the area killed the attacker with his personal firearm.

Israel didn't mandate armed guards at the entrances to all schools until 1995, the Education Ministry said ? more than two decades after the Maalot attack and two years after a Palestinian militant wounded five pupils and their principal in a knifing at a Jerusalem school.

Israel's lightly armed school guards are not the first or the last line of defense. They are backed up by special police forces on motorcycles that can be on the scene within minutes ? again bringing out the main, but not the only, difference between the two systems.

Foreign Ministry spokesman Yigal Palmor spelled it out.

"We're fighting terrorism, which comes under very specific geopolitical and military circumstances. This is not something that compares with the situation in the U.S," Palmor said.

Because it is aimed at preventing terror attacks, Israel's school security system is part of a multi-layered defense strategy that focuses on prevention and doesn't depend on a guy at a gate with a gun.

Intelligence gathering inside Palestinian territories, a large military force inside the West Bank and a barrier of towering concrete slabs and electronic fencing along and inside the West Bank provide the first line of defense.

Guards are stationed not just at schools, but at many other public facilities, including bus and train stations, parking lots, malls and restaurants.

"There are other measures of prevention of an attack taking place, which are carried out 24 hours a day, seven days a week, all over the country," police spokesman Micky Rosenfeld said. Many are not for public knowledge.

Gun lobbyists who might think Israel hands out guns freely to keep its citizens safe might be less enamored of Israel's actual gun laws, which are much stricter than those in the U.S. For one thing, notes Yakov Amit, head of the firearms licensing department at the Ministry of Public Security, Israeli law does not guarantee the right to bear arms as the U.S. Constitution does.

"The policy in Israel is restrictive," he said.

Gun licensing to private citizens is limited largely to people who are deemed to need a firearm because they work or live in dangerous areas, Amit said. West Bank settlers, for instance, can apply for weapons licenses, as can residents of communities on the borders with Lebanon and the Gaza Strip. Licensing requires multiple levels of screening, and permits must be renewed every three years. Renewal is not automatic.

The policy is designed "to strike a balance between needs and risks," Amit said. "We know that weapons are a dangerous thing, and in the hands of someone who isn't trained or isn't reliable, it causes problems."

The gap between Israeli gun ownership and U.S. gun ownership is consequently staggering. A total of 170,000 guns are licensed for private use in Israel, or about one gun for every 30 adults.

In addition to the privately held weapons, 130,000 guns are licensed to Israeli security companies, firing ranges, government ministries and companies that operate in areas deemed dangerous. Soldiers who carry assault rifles off base during their regular or reserves service turn them in when they complete their tours of duty.

By contrast, U.S. authorities estimate that at least one-third of all American households have firearms ? and in many cases, not only one.

Americans are also much freer to choose what type of guns they buy. Automatic weapons of the type Lanza used to gun down his victims are banned for private ownership in Israel. It is also rare for a person to be authorized to own more than one firearm, Amit said.

Eighty percent of the 10,000 people who apply yearly for licenses are turned down, he said. In the U.S., people can purchase firearms from private dealers without a background check or a license of any kind.

In Israel, applicants must undergo police screening and medical exams, in part to determine their mental state, Amit said.

Many Israelis receive weapons training in the military. But to be licensed to receive a weapon outside the military, they must undergo at least two hours of additional training, then repeat the training and medical exams every three years before they can renew their licenses.

Anybody who possesses a legally acquired gun waives the right to confidentiality, and authorities cross-reference for new information about the gunholder every three months.

"The point is not to complicate, but to make sure the system makes things safer," Amit said.

Businesses are increasingly conducting interviews via video ? it?s more personal than a phone interview and enables companies to interview non-local job candidates that are unable to travel.

Just like any other interview, though, there are certain things to keep in mind if you want to ace it. Adding the variable of Internet connection to the mix, Skype might even give you more to prepare for!

Here are four tips to help you rock your next Skype interview:

1. Pay attention to your background.

Speaking to an interviewer through Skype means inviting them into your environment. Not only do you want it to look good on camera, but you also want it to reflect well on yourself.

Keep it simple, but not boring.

?I would stay away from a room that?s distracting, with funky colors or gaudy wallpaper,? says Megan Morini, assistant director and business consultant at the University of Miami?s career center.

Framing a bookshelf or desk behind you is a great way to add depth to the image your interviewer will see. Just be sure to clean up all clutter and take out the trash! There?s no reason a potential employer should see Solo cups lying around.

The goal for your interview is to look professional, even though you?re at home. Be conscious of any inappropriate posters you have on the wall behind you or explicit books on your bookshelf. They may have been a gag gift from your sorority sisters, but employers have no way to know that.

While you may be tempted to interview sitting on the couch or bed, you?ll definitely want to avoid this. You?ll look far more professional sitting at a desk or table.

?What I ended up doing was going to a Barnes & Noble caf? that had free Wi-Fi to have the interview,? said Elizabeth Tompkins, a senior at the College of William and Mary. ?With that simple change of scene I felt more professional. I?m sure it looked better too, not having the background of my dorm room in the video feed.?

?I would err on the side of caution,? says Morini. ?Don?t wear anything too bright or busy. I?d stick with a black or cream-colored top.?

Don?t go halfway with your outfit, either. It can be super tempting to dress for success on the top (what can be seen on camera) and wear casual shorts or sweats on the bottom. But this could end up biting you ? you never know if you?ll have to get up at some point!

Your best bet is to dress like you would for a Monday morning at the office.

3. Study the lighting.

Your Skype interview is, essentially, your primetime moment. You?re the producer AND the star.

?Filming? conditions should be as close to perfect as you can make them so you can look like your fabulous self to your potential employer. If you position your camera in front of a window or other light source, your face will be a dark silhouette on screen. On the other hand, bright overhead or fluorescent lights can wash you out.

The best lighting is natural sunlight, but don?t worry if you?re being interviewed after dark or on a rainy day. Any light source in front of you will do.

A lamp on either side of your computer/webcam will help avoid looking like you?re under a spotlight, and you can try covering your light source(s) with a cloth to soften the light.

To test out your lighting before your interview, Morini suggests having a practice interview.

?Have a Skype call with a friend, parent or career adviser [from your campus?s career center],? she says. ?You can try out your lighting, your background, your Internet feed.?

Just like doing a dry run before an in-person interview to practice getting there, a test Skype call can really pay off.

4. Prepare for technical difficulties.

Internet connections can be spotty, so always come up with a backup plan. Skype can be plagued by technical difficulties and can cause audio lag and pixelated video ? especially when the connection is bad.

If your Wi-Fi is super unreliable, use an ethernet cable to plug in so you don?t lose service in the middle of your interview.

As much as you may love your laptop, it?s still a piece of technology and doesn?t know not to die during an interview. Keep it plugged in to avoid any potential battery disasters.

Close all unnecessary programs ? not only will they be distracting, but they?ll also slow down your computer?s processing (interfering with your video feed) and drain the battery quicker.

And make sure to keep your cell phone handy.

?Make sure you have a contact phone number for your interviewer,? Morini says. ?If something goes wrong, you can give them a call and go right into a phone interview instead.?

Chinese medicine yields secrets: Atomic mechanism of two-headed molecule derived from Chang Shan, a traditional chinese herb

Dec. 23, 2012 ? The mysterious inner workings of Chang Shan -- a Chinese herbal medicine used for thousands of years to treat fevers associated with malaria -- have been uncovered thanks to a high-resolution structure solved at The Scripps Research Institute (TSRI).

Described in the journal Nature this week, the structure shows in atomic detail how a two-headed compound derived from the active ingredient in Chang Shan works. Scientists have known that this compound, called halofuginone (a derivative of the febrifugine), can suppress parts of the immune system -- but nobody knew exactly how.

The new structure shows that, like a wrench in the works, halofuginone jams the gears of a molecular machine that carries out "aminoacylation," a crucial biological process that allows organisms to synthesize the proteins they need to live. Chang Shan, also known as Dichroa febrifuga Lour, probably helps with malarial fevers because traces of a halofuginone-like chemical in the herb interfere with this same process in malaria parasites, killing them in an infected person's bloodstream.

"Our new results solved a mystery that has puzzled people about the mechanism of action of a medicine that has been used to treat fever from a malaria infection going back probably 2,000 years or more," said Paul Schimmel, PhD, the Ernest and Jean Hahn Professor and Chair of Molecular Biology and Chemistry and member of The Skaggs Institute for Chemical Biology at TSRI. Schimmel led the research with TSRI postdoctoral fellow Huihao Zhou, PhD.

Halofuginone has been in clinical trials for cancer, but the high-resolution picture of the molecule suggests it has a modularity that would make it useful as a template to create new drugs for numerous other diseases.

The Process of Aminoacylation and its Importance to Life

Aminoacylation is a crucial step in the synthesis of proteins, the end products of gene expression. When genes are expressed, their DNA sequence is first read and transcribed into RNA, a similar molecule. The RNA is then translated into proteins, which are chemically very different from DNA and RNA but are composed of chains of amino acid molecules strung together in the order called for in the DNA.

Necessary for this translation process are a set of molecules known as transfer RNAs (tRNAs), which shuttle amino acids to the growing protein chain where they are added like pearls on a string. But before the tRNAs can move the pearls in place, they must first grab hold of them.

Aminoacylation is the biological process whereby the amino acid's pearls are attached to these tRNA shuttles. A class of enzymes known as aminoacyl-tRNA synthetases is responsible for attaching the amino acids to the tRNAs, and Schimmel and his colleagues have been examining the molecular details of this process for years. Their work has given scientists insight into everything from early evolution to possible targets for future drug development.

Over time what has emerged as the picture of this process basically involves three molecular players: a tRNA, an amino acid and the aminoacyl-tRNA synthetase enzyme that brings them together. A fourth molecule called ATP is a microscopic form of fuel that gets consumed in the process.

The new work shows that halofuginone gets its potency by interfering with the tRNA synthetase enzyme that attaches the amino acid proline to the appropriate tRNA. It does this by blocking the active site of the enzyme where both the tRNA and the amino acid come together, with each half of the halofuginone blocking one side or the other.

Interestingly, said Schimmel, ATP is also needed for the halofuginone to bind. Nothing like that has ever been seen in biochemistry before.

"This is a remarkable example where a substrate of an enzyme (ATP) captures an inhibitor of the same enzyme, so that you have an enzyme-substrate-inhibitor complex," said Schimmel.

Dear EarthTalk: What is ?perchlorate? in our drinking water supply, and why is it controversial??David Sparrow, Chico, Calif.

Perchlorate is both a naturally occurring and man-made chemical used in the production of rocket fuel, missiles, fireworks, flares and explosives. It is also sometimes present in bleach and in some fertilizers. Its widespread release into the environment is primarily associated with defense contracting, military operations and aerospace programs.

Perchlorate can be widespread in ground water, soils and plants, and makes its way up the food chain accordingly?even into organically grown foods. To wit, A 2005 Journal of Environmental Science and Technology study using ion chromatography to find contaminants in agricultural products found quantifiable levels of perchlorate in 16 percent of conventionally produced lettuces and other leafy greens and in 32 percent of otherwise similar but organically produced samples. Today, traces of perchlorate are found in the bloodstreams of just about every human on the planet.

Perchlorate in the environment is a health concern because it can disrupt the thyroid?s ability to produce hormones needed for normal growth and development. Besides its potential to cause endocrine system and reproductive problems, perchlorate is considered a ?likely human carcinogen? by the U.S. Environmental Protection Agency (EPA). Some 11 million Americans live in areas where concentrations of perchlorate in public drinking water supplies are significantly higher than what is considered safe.

Per the mandate of the Safe Drinking Water Act, the EPA is currently working on setting national standards for how much perchlorate can be allowed in drinking water without putting people at risk. As part of the process, the agency is studying the available science on the health effects of perchlorate exposure and evaluating laboratory methods for measuring, treating and removing perchlorate in drinking water. The EPA will publish a proposed rule on the matter for public review at some point in 2013.

?We are happy that the EPA is moving ahead with a drinking water standard...but we are concerned that it won?t be strict enough,? reports Renee Sharp of the nonprofit Environmental Working Group (EWG). The group would like to see the U.S. adopt ?a truly health-protective drinking water standard lower than 1 ppb [parts per billion]? for perchlorate. Insiders don?t believe federal policymakers will go that low, however, since the EPA says it cannot detect perchlorate below 2 ppb. But EWG point out that Massachusetts is already testing for it with a 1 ppb cut-off, per the mandate of its statewide standard set back in 2006.

The only other state to have a drinking water standard for perchlorate is California, which set 6 ppb or less as an allowable concentration back in 2004. But that state?s Office of Environmental Health Hazard Assessment recently proposed lowering the standard to 1 ppb based on new data regarding environmental exposure, possible effects of perchlorate and consideration of infants as a susceptible population.

If the EPA develops a tough new standard, almost every state will need to readjust its water monitoring systems to take into account how much perchlorate is making its way to our taps and into the foods we eat?a no doubt costly process but one that will greatly benefit both current and? future generations.

The award will be presented at the WGAW?s 2013 Writers Guild Awards West Coast ceremony on Sunday, February 17, 2013, at the JW Marriott Los Angeles L.A. LIVE.

?Dan Petrie, Jr. is a remarkable man, distinguished for his creative accomplishments as an Oscar?-nominated screenwriter, as a producer and as a director, and no less distinguished for his long and passionate commitment to the well-being of all writers. Dan has served this Guild for 25 years, twice as President. He has guided us through difficult times. A champion of our fervent and often raucous democracy, with a quiet manner and a firm hand, Dan made our many voices our strength. In the process, he earned the respect of all who served with him. He has always had mine. It is fitting, therefore, that Dan Petrie, Jr. should be the unanimous choice of the Board to receive this year?s Morgan Cox Award,? said WGAW President Christopher Keyser.

Screenwriter-director-producer Petrie was nominated for an Academy Award for his first produced script, the box-office hit Beverly Hills Cop (1984), starring Eddie Murphy (Screenplay by Daniel Petrie, Jr., Story by Danilo Bach and Daniel Petrie, Jr.). He followed up this success with his original screenplay for the romantic thriller, The Big Easy (1986), which received an Edgar Allen Poe Award nomination for Best Motion Picture and was later adapted into a 1996-97 TV series. Petrie then served as producer of the thriller Shoot to Kill (1988), which he co-wrote (Screenplay by Harv Zimmel and Michael Burton and Daniel Petrie, Jr., Story by Harv Zimmel), and as executive producer of the comedy Turner & Hooch (1989), starring Tom Hanks, which he co-wrote (Screenplay by Dennis Shryack & Michael Blodgett and Daniel Petrie, Jr. and Jim Cash & Jack Epps, Jr., Story by Dennis Shryack & Michael Blodgett).

Petrie made his directorial debut with the 1991 action-adventure feature Toy Soldiers, which he co-wrote (Screenplay by Daniel Petrie, Jr. and David Koepp, Based on the novel by William Kennedy). Petrie also directed the HBO film Dead Silence, as well as adapted and directed the TNT movie Framed. Petrie also co-wrote the acclaimed Hallmark Hall of Fame telefilm Pictures of Hollis Woods (Teleplay by Ann Peacock and Daniel Petrie, Jr. and Camille Thomasson, Based on the Novel by Patricia Reilly Giff), which earned the trio a Humanitas Prize nomination (90 Minute or Longer Network or Syndicated Television). Most recently, Petrie was executive producer, showrunner, and co-creator of the military medical drama Combat Hospital (Created by Jinder Oujila-Chalmers & Douglas Steinberg and Daniel Petrie, Jr.), a 13-episode series which was simulcast on Canada?s Global TV and ABC in the U.S. during the summer of 2011.

In 2006, Petrie and producing partner Rick Dugdale founded Enderby Entertainment, an independent film, television, and digital media company with an old-fashioned emphasis on storytelling on the creative side, and transparency on the financial side. The company recently arranged the financing and co-produced writer-director Stephen Elliot?s film About Cherry, which premiered at the 2012 Berlin International Film Festival. Enderby?s thriller division, Tony-Seven Films, has also financed and produced six genre pictures to date: The Speak, Vile, 5 Souls, Silver Falls, No Tell Motel, and Bloodshed.

A Writers Guild member since 1984, Petrie has an extensive history of service to the Guild, including serving two terms as WGAW President (1997-99, 2004-05) and three terms as Vice President (1995-97, 1999-01, 2004), as well as a member of the WGAW Board of Directors (1994-95). Petrie currently serves as the Writers Guild Foundation?s Vice President of Programs, having held that position since 2009. His other Foundation service includes stints on the WGF?s Board of Directors (1995-03, 2005-08), as Treasurer (2008), and as Vice President of Development (2001-04). Petrie inherited a tradition of volunteer service to the motion picture and television industry from his parents: his father was the late Canadian director and longtime Directors Guild of America (DGA) Board member Daniel Petrie, Sr., while mother is producer and longtime Producers Guild of America (PGA) Board member Dorothea Petrie. Petrie lives in Los Angeles with his wife, Constance Petrie.

The Morgan Cox Award honors that WGAW member whose vital ideas, continuing efforts, and personal sacrifice best exemplify the ideal of service to the Guild. Previous recipients include George Kirgo, Del Reisman, Frank Pierson, Carl Gottlieb, and Patric M. Verrone. During his lifetime, the late Cox devoted his professional career to serving the Guild, working to ensure that television writers were included under WGA jurisdiction.
